目的 探索利用升高透明平台法建立动物焦虑行为的效果和咪达唑仑对减轻利用升高透明平台法诱导的小鼠焦虑行为的作用。
方法 雄性C57BL/6J小鼠39只,2月龄,体重20~24 g。将小鼠随机分为三组:对照组(C组)、升高平台应激组(E组)和升高平台应激+咪达唑仑组(EM组),每组13只。C组正常饲养;E组每天接受2次(每次持续3 min)升高透明平台应激,连续1周;EM组小鼠除接受与E组相同的升高透明平台应激外,于建模结束后第1天和第3天的行为学测试前30 min腹腔注射咪达唑仑0.5 mg/kg。建模结束后,采用旷场实验与新环境进食抑制实验检测小鼠进入旷场中心区次数、中心区停留时间、中心区运动距离和进食潜伏期。采用ELISA法检测小鼠血清皮质酮(Cor)浓度及海马组织中γ-氨基丁酸(GABA)浓度。
结果 与C组比较,E组中心区进入次数明显减少(P<0.05)、中心区停留时间和中心区运动距离明显缩短(P<0.05),进食潜伏期明显延长(P<0.05),血清Cor浓度明显升高(P<0.05),海马组织GABA浓度明显降低(P<0.05)。与E组比较,EM组中心区进入次数明显增加(P<0.05),中心区时间明显延长(P<0.05),进食潜伏期明显缩短(P<0.05),血清Cor浓度明显降低(P<0.05),海马组织中GABA浓度明显升高(P<0.05)。
结论 升高透明平台应激可诱导小鼠产生焦虑样行为,咪达唑仑可改善小鼠焦虑样行为,降低血清Cor浓度,升高海马组织GABA浓度。

Objective To explore the effect of establishing anxiety behavior in animals by using elevated transparent platform method and the effect of midazolam on alleviating anxiety behavior in mice induced by elevated transparent platform method.
Methods Thirty-nine male C57BL/6J mice, aged 2 months, weighing 20-24 g, were randomly divided into three groups: control group (group C), elevated platform stress group (group E), and elevated platform stress + midazolam group (group EM), 13 mice in each group. Mice in group C were fed normally, mice in group E received elevated transparent platform stimulation twice a day (lasted for 3 minutes) for a week. In group EM, midazolam 0.5 mg/kg was intraperitoneally injected 30 minutes before the behavioral test on the first and third days after modeling, in addition to the stimulation of the elevated transparent platform. After the completion of the three models, open field experiment and new environment eating inhibition experiment were used to detect the frequency of entering the central area of the open field, the central area time, the movement distance and the feeding latency of the mice. Serum corticosterone (Cor) concentration and γ-aminobutyric acid (GABA) concentration in hippocampus of mice were determined by ELISA.
Results Compared with group C, the frequency, time and movement distance of mice entering the central area of the open field in group E were significantly reduced (P < 0.05), eating in a new environment was significantly prolong the incubation period (P < 0.05), Cor concentration was significantly increased (P < 0.05), GABA concentration in hippocampus was significantly decreased (P < 0.05). Compared with group E, the frequency and time of entering the central area of open field in group EM was significantly increased (P < 0.05), the incubation period of eating in new environment was significantly shortened (P < 0.05), the concentration of Cor was significantly decreased (P < 0.05), and the concentration of GABA in hippocampus was significantly increased (P < 0.05).
Conclusion Elevated transparent platform stress can successfully induce anxiety-like behavior in mice, while midazolam improved anxious-like behavior, decreased serum Cor concentration and increased hippocampal GABA concentration.
