目的 探讨氯胺酮对新生大鼠海马区G蛋白偶联受体30(G protein-coupled receptor 30, GPR30)表达的影响。
方法 实验一:随机选取20只健康雄性SD大鼠作为空白对照组(S组,n=20),在饲养1、3、7、14 d时各取出5只处死,采用免疫组化法检测大鼠海马区GPR30阳性细胞数。实验二:10只出生7 d健康雄性SD大鼠随机分为两组,生理盐水组(C组,n=5)和氯胺酮组(K组,n=5)。连续3 d腹腔注射生理盐水0.1 ml(C组)或氯胺酮75 mg/kg(K组),在末次给药后24 h处死,采用免疫组化法检测大鼠海马区cleaved caspase-3阳性细胞数,采用Western blot法检测海马区GPR30蛋白含量。
结果 与1 d和3 d比较,7 d和14 d S组大鼠海马区GPR30蛋白含量明显升高(P<0.05);与7 d比较,14 d时S组大鼠海马区GPR30蛋白含量明显升高(P<0.05)。与C组比较,K组海马区cleaved caspase-3阳性细胞数明显增多,GPR30蛋白含量明显降低(P<0.05)。
结论 氯胺酮诱导发育期大鼠海马细胞凋亡可能与GPR30表达下调有关。

Ojective To investigate the effect of ketamine on the expression of G protein-coupled receptor 30 (GPR30) in hippocampus of neonatal rats.
Methods Step 1: 20 healthy male Sprague-Dawley rats were randomly selected as blank control group (group S, n = 20), five rats were taken out at 1, 3, 7 and 14 days of feeding, the number of positive cells of GPR30 in the hippocampus of rats was detected by immunohistochemistry. Step 2: 10 7-day-old healthy male Sprague-Dawley rats were randomly divided into two groups: saline group (group C, n = 5) and ketamine group (group K, n = 5). Three days after intravenous injection of 0.1 ml (group C) or ketamine 75 mg/kg (group K), the rats were sacrificed at 24 h after the last administration. The expression of cleaved caspase-3 in hippocampus of rats was detected by immunohistochemistry and the expression of GPR30 in hippocampus was detected by Western blot.
Results Compared with 1 d and 3 d of group S, the expression of GPR30 in hippocampus of 7 d and 14 d rats was significantly increased (P < 0.05). Compared with 7 d of group S, the expression of GPR30 in hippocampus of 14 d rats was significantly increased (P < 0.05). The levels of cleaved caspase-3 in hippocampus were significantly higher and the levels of GPR30 in hippocampus were significantly lower in group K than in group C (P < 0.05).
Conclusion Ketamine-induces hippocampus cell apoptosis in the developing rat brain, which is associated with the decrease level of GPR30.
