目的 观察临床常用的静脉麻醉药丙泊酚对大鼠离体完整海马CA1区场电位不同振荡频段的作用,探讨丙泊酚对海马内部神经环路功能可能产生的影响。
方法 选择出生后14~16 d雄性SD大鼠于4℃条件下进行完整海马的剥离。待孵育1~2 h稳定后,移至电生理记录槽。实验根据灌流的丙泊酚浓度不同分为对照组(C组)、丙泊酚1 μmol/L组(P1组)、丙泊酚20 μmol/L组(P20组)、丙泊酚40 μmol/L组(P40组)和丙泊酚80 μmol/L组(P80组)。C组不予丙泊酚灌流,为基础值,P1组、P20组、P40组和P80组分别给予丙泊酚1、20、40和80 μmol/L灌流,采用玻璃微电极记录离体完整海马CA1区辐射层的场电位信号,进行场电位的功率谱强度分析。将C组的基础值设定为1,分别计算并比较P1组、P20组、P40组和P80组离体完整海马CA1区场电位不同振荡频段(δ:1~4 Hz,θ:4~8 Hz,α:8~13 Hz,β:13~30 Hz,γ:30~80 Hz)与C组比值。
结果 与C组比较,P1组场电位θ频段明显增强(P<0.05),γ频段明显减弱(P<0.05),P20组、P40组和P80组场电位不同振荡频段均明显减弱(P<0.05)。与P1组比较,P20组场电位δ、θ、α、β频段明显减弱(P<0.05),P40组和P80组场电位不同振荡频段均明显减弱(P<0.05)。与P20组比较,P80组场电位δ、θ、α频段明显减弱(P<0.05)。与P40组比较,P80组场电位δ频段明显减弱(P<0.05)。
结论 除θ频段外,丙泊酚对海马CA1区场电位的δ、α、β和γ频段呈现浓度依赖的抑制作用;对于θ频段,大于20 μmol/L浓度的丙泊酚仍然表现为抑制作用,但在1 μmol/L浓度下却表现为增强作用,显示低浓度丙泊酚对海马内部神经环路可能存在兴奋作用。

Ojective To reveal the effect of propofol on hippocampal neuroal circuits, local field potential (LFP) in CA1 region of intact hippocampus in vitro.
Methods SD rats aged 14 - 16 days postnatal were anesthetized and decapitated. The hippocampal formation was dissected from brain tissue in 4℃. LFP of hippocampal CA1 stratum radiatum was recorded with glass electrode when the bath was perfused with propofol at 5 different concentrations-control group with no propofol (group C), 1 μmol/L (group P1), 20 μmol/L(group P20), 40 μmol/L (group P40) and 80 μmol/L (group P80), respectively. The effect of propofol on the LFP power spectrum (δ: 1~4 Hz, θ: 4~8 Hz, α: 8~13 Hz, β: 13~30 Hz, γ: 30~80 Hz) in CA1 region of intact hippocampus was analyzed.
Results Compared with group C, θ oscillation in group P1 was significantly increased (P < 0.05), γ oscillation was significantly decreased (P < 0.05), while all oscillations of LFP power spectrum were obviously decreased in groups P20, P40 and P80(P < 0.05). Compared with group P1, δ, θ, α and β oscillations in group P20 were significantly decreased (P < 0.05). And all oscillations of LFP power spectrum in groups P40 and P80 were significantly decreased (P < 0.05). Compared with group P20, δ、θ、α oscillations in groups P40 were significantly decreased (P < 0.05). Compared with group P40, δ oscillation in group P80 was significantly decreased (P < 0.05).
Conclusion Propofol has a concentration-dependent inhibitory effect on δ, α, β and γ oscillations of hippocampal CA1. Although propofol at over 20 μmol/L inhibits θ oscillation, propofol at 1 μmol/L enhances θ oscillation, suggesting that propofol at low concentration exhibits exciting effects on the hippocampal internal neural circuits.
