目的 观察腺苷A1受体在右美托咪定调节压力反射敏感性(baroreflex sensitivity, BRS)中的作用。

方法 健康成年雄性SD大鼠32只, 体重240~280 g, 按随机数字表随机分为四组: 对照组(C组)、选择性腺苷A1受体阻断剂组(P组)、右美托咪定组(D组)、选择性腺苷A1受体阻断剂+右美托咪定组(PD组), 每组8只。C组泵注生理盐水40 ml·kg-1·h-1负荷量15 min,维持泵注10 ml·kg-1·h-1;P组腹腔注射选择性腺苷A1受体阻断剂8-环戊基-1, 3-二丙基黄嘌呤(DPCPX)1 mg/kg, 泵注同C组方案的生理盐水;D组右美托咪定负荷量100 μg/kg, 维持量100 μg·kg-1·h-1持续泵注;PD组腹腔注射DPCPX 1 mg/kg并泵注右美托咪定, 泵注剂量同D组。采用苯肾上腺素升压法于泵注前(T0)、泵注后60 min(T1)和泵注后120 min(T2)测定BRS。

结果 与T0时比较, T1和T2时D组和PD组BRS明显升高(P<0.05)。与C组和P组比较, T1和T2时D组和PD组BRS均明显升高(P<0.05)。与D组比较, T1和T2时PD组BRS明显降低(P<0.05)。

结论 右美托咪定可能通过腺苷A1受体增加大鼠BRS。

Objective To investigate the role of adenosine A1 receptor in the regulation of baroreflex sensitivity (BRS) by dexmedetomidine in rats.

Methods Thirty-two healthy male Sprague-Dawley rats, weighting 240 - 280 g, were randomly divided into 4 groups, with 8 rats in each group. The equal volume of normal saline was administered in group C as compared to expermental groups. Intraperitoneal injection of selective adenosine A1 receptor blocker 8-cyclopentyl-1, 3-dipropylxanthine (DPCPX) 1 mg/kg was used first, and then the equal volume of normal saline was administered in group P. Dexmedetomidine 100 μg/kg was administered within 15 minutes followed by infusion at 100 μg·kg-1·h-1 in group D. Intraperitoneal injection of DPCPX 1 mg/kg was used first, and then dexmedetomidine 100 μg/kg was administered within 15 minutes followed by infusion at 100 μg·kg-1·h-1 in group PD. BRS was measured before (T0), 60 min after (T1) and 120 min after (T2) intravenous pumping of dexmedetomidine or normal saline.

Results Compared with T0, the BRS was significantly higher in groups D and PD at T1 and T2 (P < 0.05). Compared with groups C and P, BRS was significantly higher in groups D and PD at T1 and T2 (P < 0.05). Compared with group D, the BRS was significantly lower in group PD at T1 and T2 (P < 0.05).

Conclusion Dexmedetomidine can increase the BRS of rats with adenosine A1 receptor.
