| 慢性疼痛是一种临床高发的顽固性疾病,但目前尚无显效的治疗措施,明确其发病机理一直是医学研究的重要方向。细胞程序性坏死(Nec)是一种新发现的可调控性细胞死亡模式,最新研究表明,Nec可通过不同的分子机制促使慢性疼痛的发生和发展,条件性敲除或给予靶向药物抑制Nec关键通路蛋白的活性均可显著改善动物模型所形成的疼痛异常行为,故Nec可能是一种有效治疗慢性疼痛的新突破口。基于上述背景,本文针对Nec的激活途径、在慢性疼痛形成中的作用以及可能的分子机制进行重点论述。 |

| Chronic pain is a refractory disease with a high clinical incidence, yet effective treatment measures remain elusive. Consequently, investigating the pathogenesis of chronic pain has consistently been a focal point of medical research. Recent studies have identified necroptosis as a novel regulatory cell death mode, which has been shown to facilitate the onset and progression of chronic pain through various molecular mechanisms. Notably, the inhibition of key necroptosis proteins through knock out or targeted drug interventions has demonstrated significant improvements in abnormal behavior within animal models. Thus, necroptosis holds promising potential as a novel therapeutic avenue for the treatment of chronic pain. In light of the above background, this article focuses on the activation pathway of necroptosis, its role and possible molecular mechanisms in chronic pain. |
